1.5.6 Calculating the Frequency of Nucleotide Bases

infoNote
  • DNA is composed of four nucleotide bases: adenine (A), thymine (T), cytosine (C), and guanine (G).
  • These bases pair specifically:
  • Adenine pairs with Thymine (A-T)
  • Cytosine pairs with Guanine (C-G)
  • The proportion of one base affects the proportions of its complementary base due to these pairing rules.

Steps for Calculating Nucleotide Base Frequencies

  1. Understand Base Pairing Rules:
  • The percentage of adenine (A) equals the percentage of thymine (T).
  • The percentage of cytosine (C) equals the percentage of guanine (G).
  • Thus, if you know the percentage of one base, you can calculate the others.
  1. Using a Known Percentage:
  • If A=x%A = x\%, then T=x%T = x\%.
  • Similarly, C+G=100%(A+T)C + G = 100\% - (A + T).
  • If C=y%C = y\%, then G=y%.G = y\%.
  1. Example Calculation:
  • Suppose A=:highlight[30%]A = :highlight[30\%].
  • T must also be :highlight[30%](A=T):highlight[30\%] (A = T).
  • Therefore, C+G=100%(A+T)=100%60%=:highlight[40%]C + G = 100\% - (A + T) = 100\% - 60\% = :highlight[40\%].
  • If C=:highlight[20%]C = :highlight[20\%], then G=:highlight[20%]G = :highlight[20\%].
infoNote

Exam Tips

  • Always check the question for additional context about base proportions.

  • If given a double-stranded DNA sequence, remember that the total percentages of A, T, C, and G must equal :highlight[100%].:highlight[100\%].

  • Double-check your arithmetic when determining complementary percentages.

  • Be familiar with questions asking for one base when others are given.

infoNote

Key Formula:

C+G=100%(A+T)C + G=100\%−(A + T)
